SDS PAGE Gel

Chemical ingredients
-> Tris : used as a buffer, innocuous substance to most protein
-> Glycine : used as the source of trailing ion or slow ion
-> Acrylamide
-> Bisacrylamide : frequently used cross linking agent
-> Sodium Dodecyl Sulfate (SDS) : dissociation agent, denature native protein => cause native protein to unfold
-> Ammonium persulfate (APS) : initiator for gel formation
-> TEMED

Method
각각 조성에 맞게 Running gel 과 Stacking gel을 만든다
이때 TEMED는 나중에 넣는다 (TEMED를 넣는 순간부터 gel 화 되기 때문이다)
70% alcohol로 short plate 와 plate 를 먼지가 없도록 잘 닦는다
gasket (고무같은) 을 frame 밑에 넣고 닦은 유리를 frame에 넣는다
Stacking gel 과 Running gel 을 short plate 와 plate 사이에 넣어야 하는데
Running gel 이 밑에 있으므로 먼저 넣는다
이때 기포가 발생 할 수도 있는데, 에탄올을 넣으면 해결된다. 또한 에탄올을 넣게 되면 공기도 차단된다.
15분 후, 프레임을 잡고 뒤집어 에탄올을 뺀다. 아까 넣은 gel은 굳어서 빠지지 않는다.
남은 에탄올은 filter paper 로 흡수시킨다.
TEMED 를 넣은 Stacking gel을 틀에 넣고 comb를 끼운다.
15분 후, 흐르는 물에서 comb을 뺀다.
틀에서 유리를 분리 시키고 랩에 싸서 4'C에 보관한다ㅇㅇㅇㅇㅇ
